This study was conducted to evaluate whether Rhodiola sachalinesis extract (RS) is effective in reducing Saw palmetto extract (SP)-induced toxicity and improving benign prostatic hyperplasia (BPH). RS powder was extracted with 50% ethanol to prepare an extract. The 2, 2-diphenyl-1-picrylhydrazyl (DPPH) and 2, 2-diphenyl-1-picrylhydrazyl (ABTS) radical scavenging activities for RS extract were 8.7 ¥ìg/mL and 2.5 ¥ìg/mL, respectively. Toxicity to testicular cells, normal prostate cells and BPH-1 by SP or RS alone treatment and SP-RS combined treatment was tested. As a result, SP showed toxicity in testicular cells and normal prostate cells, whereas RS did not show toxicity in both cells. On the other hand, SP did not show apoptotic effect in BPH-1, but RS-induced apoptosis in BPH-1 in a concentration-dependent manner. In SP-RS complex treatment, the normal intracellular toxicity to SP was weakened, and the cytotoxic effect on BPH-1 was rather increased. These results indicate that RS can be used as a material for BPH improvement because it can induce apoptosis of BPH-1 without toxicity to normal cells, when treated with SP alone or in combination with SP.
